Melanoma cells are auxotrophic for arginine, because they lack argininosuccinatesynthetase (ASS), a key enzyme required for the synthesis of arginine. WebADI is a metabolizing enzyme converting Arg into citrulline and was first extracted from Mycoplasma [ 27 ]. Because of its instability and rapid clearance from the plasma, a more stabilized form of this enzyme called ADI- polyethylene glycol 20 (ADI-PEG20) has been synthesized for therapy [ 28 ].
